Sorting of Perkinsus with high salt concentration culture media

[Summary]
Cloning of Perkinsus which is a kind of protozoa and lives on oysters was done with high salt concentration artificial sea water. It can be done without damage because it does not have to replace sea water to common sheath liquid.

[Introduction]
The example of an experiment for Perkinsus using On-chip Sort and sea water showed that a cell sorter is an effective method to the cloning of the many kind of cells.

[Method]
The solution which diluted Through Path Plus (Cat# 2001004-6 Lot# 131018) with the high salt concentration culture medium (40 times) is used for sheath liquid, Sample liquid used the solution which mixed an equivalent amount of high salt concentration culture media and Sample buffer (Cat# 2001005 Lot# 130315).

[Result]
With On-chip Sort, we could sort Perkinsus easily using culture media which was made from high salt concentration artificial sea water. The damage of Perkinsus was not observed because we did not change the culture media to another one such as PBS and so on. On-chip Sort is an extremely useful tool when you sort some cells like Perkinsus which are in high salt concentration culture media because On-chip Sort does not have limitation about sheath. If you utilize its peculiar character, you can clone cells quickly and easily.
