【Summary】 The nerve cells were adjusted from the rat fetal brain hippocampus, and the influence of the damage which sorting gives to formation of nerve axon were considered. As for the nerve cells sorted by “On-chip Sort”, formation of the nerve axon was observed after sorting. However, the nerve cells sorted with the existing cell sorter became extinct without to forming nerve axon during the cultivation after sorting.
【Purpose】 The nerve cells were prepared from the rat fetal brain hippocampus, and the influence of the damage which sorting gives to formation of nerve axon were considered.
【Method】 The neuronal cells were obtained from rat fetal brain hippocampus, and three classifications of “without sorting”, “the cells sorted with conventional sorter of high-speed Jet-in-Air system” and “the cells sorted using On-chip Sort” was prepared.
The percentage of dead cells was seen from the rate of the cell dyed with PI.
【Result 1】 The survival rate of control (without sorting) was 83%, sorted cell with the existing sorter was 77% , sorted cell by “On-chip Sort” was 80%.
【Result 2】 As for the nerve cells which sorted in “On-chip Sort”, the nerve axonal formation was observed 3 to 7days after sorting, but the nerve cells which sorted with an existing cell sorter perished without being able to form a nerve axon in the culture after sorting.
