Comparison between On-chip Sort and existing cell sorter -Eosinophil sorting and observation in the morphology

【Summary】   On-chip Sort does not have damage to give a cell in comparison with the cell sorter of the conventional Jet-in-Air method. It was thought that the peripheral blood mononuclear leukocyte resisted comparative damage, but the cell which sorted in On-chip Sort kept a form of normal eosinophil, but the cell which sorted with the sorter of the droplet generation method was not able to keep the form of eosinophil.

【Purpose】  We compared the influence to give a cell shape when we sorted a peripheral blood cell in both On-chip technology and high-speed Jet-in-Air method, and the damage which sorting gives to a cell was examined.

【Method】  The cell of CD193 and CD123 positivity was sorted from human peripheral blood, and the form of the cell was observed.

【Result】   The cell which sorted in On-chip Sort kept a form peculiar to eosinophil, but the cell which sorted with the sorter of the droplet generation method was not able to keep the form of the normal cell. It became clear that the damage to give a cell was heavy so that the diameter of the nozzle was small with the sorter of the droplet generation method.
